Despite the absence of injured skipper Meg Lanning, Australia are in the box seat to claim a record-equalling 21 straight ODI wins after posting 5-325 at Allan Border Field.
It is their highest ODI total against the White Ferns, their second biggest on Australian soil and fourth best overall.
New Zealand will have to re-write the record books to win.
Australia's 6-289 against the White Ferns in 2012 is the highest run chase in women's ODI history.
Stand-in captain Rachael Haynes (96 off 104 balls) led the way on Wednesday, stepping up for the absent Lanning.
Lanning suffered a hamstring injury during her match-winning, unbeaten century in Australia's four-wicket game two victory on Monday that sealed the Rose Bowl trophy.
Australia now aim to equal the record of 21 consecutive victories set by Ricky Ponting's 2003 side.
Haynes enjoyed a chance on 81 to post her 14th ODI half century, hitting 10 fours and two sixes to just fall short of her second limited-overs ton.
She shared a 144-run opening stand with Alyssa Healy who rode her luck in a run-a-ball 87 featuring 13 fours and a six.
Healy notched her highest ODI score against the White Ferns after having chances on 61 and 67.
New Zealand leg-spinner Amelia Kerr took 3-50 off 10 overs.
Australia had big shoes to fill without Lanning who had not been dismissed this series after scores of 62 not out and an unbeaten 101.
"It is a slight strain. I took one of Rachael Haynes' quick singles (on Monday) and didn't come out of it too good," Lanning said when asked about her injury.
"It's nothing too serious. Hopefully I will be back fairly soon."
Lanning was replaced in the line-up by Tahlia McGrath, marking the all-rounder's first game for Australia since the 2017 Ashes.
New Zealand's attack featured Holly Huddleston, who replaced Lea Tahuhu (back/side strain).
