About 3.30am on July 16, the Westpac Rescue Helicopter Service (WRHS) was tasked on a primary mission to Wingen, along with other emergency units in the Upper Hunter.
However, the chopper then rendezvoused with two road ambulances at the Muswellbrook District Hospital's helipad.
The WRHS transported a 21-year-old female and a man, 30, who had suffered injuries sustained in the accident, to John Hunter Hospital.
They were deemed to be in a stable condition, after receiving treatment from local NSW Ambulance paramedics and the service's critical care medical team.
A third patient was taken by ambulance to an unknown hospital.
There are no further details.
